This is an expression assay — it measures how much protein each construct produced. It is not a measurement of cell penetration, biological activity, target binding, or therapeutic efficacy.
What was observed
First returned wet-lab package: a SingleChain expression screen reporting per-construct protein yield for four CPP-panel constructs (two Protean CPP candidates + two negative controls). Three of four constructs expressed above the assay's upper quantification limit; one candidate measured 91.8 ug/mL. This is experimental evidence for the measured expression endpoint only — not cell penetration, biological activity, target binding, developability beyond yield, or clinical/therapeutic efficacy.

Two constructs are labelled '-neg' (negative). Their intended role and expected behaviour are NOT documented in the package, and both express at the highest reported yields. Their sequences are NOT Protean candidates. Control behaviour therefore cannot be validated from this package.

- Cell penetration / cellular uptake (not measured).
- Biological activity, target binding, or efficacy (targetless; not measured).
- Developability beyond expression yield (no solubility/purity/aggregation).
- A unique Protean candidate_id linkage (sequence maps one-to-many).
- Control validation (negative-control semantics + behaviour indeterminate).
- Any comparative ranking (values are censored).
Next recommended experiment
A cellular-uptake / cell-penetration assay on the expressed candidates would be required before any cell-penetration claim; expression establishes only that the protein can be produced.
Limitations
- Experimental evidence for the measured expression endpoint only — not a claim of cell penetration, biological activity, target binding, developability beyond yield, or clinical/therapeutic efficacy.
- Three of four constructs expressed above the assay's upper quantification limit (reported as '>value'), so those values are lower bounds and no comparative ranking is possible.
